Pneumococcus Antisera for Neufeld

Pneumococci are enclosed within a capsule consisting of repeated units of polysaccharides. The composition of the polysaccharides defines the serotype, and 91 pneumococcal serotypes have been described to date, with more on the way. The 91 serotypes are divided into 25 types and 21 groups. Each group consists of 2-5 serotypes which are very similar.

Product description

Pool, type and factor antisera are produced by immunising rabbits with killed whole cells. Pool antisera are a mixture of antisera. Pools A-I contain antisera against all serotypes, while pools P‑T are mixtures of antisera against the 23-valent vaccine serotypes. By combining the two Pool systems (the Pneumotest products), the 23 serotypes can be serotyped easily to type/group level.

Contains antisera against

Pool A, type 1, 2, 4, 5, group 18
Pool B, type 3, 8, group 6, 19
Pool C, type 20, 31, 40, group 7, 24
Pool D, type 36, 37, group 9, 11, 16
Pool E, type 21, 39, group 10, 12, 33
Pool F, type 27, group 17, 22, 32, 41
Pool G, type 29, 34, 42, group 35, 47
Pool H, type 13, 14, group 15, 23, 28
Pool I, type 38, 43, 44, 45, 46, 48, group 25
Pool P, type 1, 14, group 7, 19
Pool Q, group 6, 18, 23
Pool R, type 3, 4, group 9, 12
Pool S, type 5, 8, group 10, 15, 17
Pool T, type 2, 20, group 11, 22, 33

Type antisera react specifically with the serotype in question.
Group antisera are specific antisera reacting with all serotypes in the group in question. The ultimate identification of the serotypes in the group is carried out by serotyping with factor antisera. The combination of the results defines the serotype.
All the products are absorbed free of cross-reactions, but factor sera are absorbed free of cross‑reactions only within the group. In the individual cases below, it is not possible to produce a specific antiserum without losing titre of the homologous antibody. Therefore, the following cross-reactions will occur:

Antiserum cross-reacting type

Group 25: 38
Type 29: 35B
Group 35: 42, 47F
Type 42: 20, 31 33A, 35A, 35B, 35C
Each product is supplied in a vial with 1 ml ready-to-use antiserum, sufficient for approx. 300 tests.

How to use

Pool, type, group and factor antisera are used in capsular reactions, also called the Neufeld test. Equal quantities of antiserum and bacterial culture are mixed on a slide. Under a phase contrast microscope, a positive reaction is indicated when the capsule enclosing the pneumococcus swells and becomes visible. The bacteria will frequently also agglutinate.

Product range

The 14 pools can be bought individually, or 12 of them can be bought together in the Pneumotest-kit. There are 25 type antisera, 21 group antisera and 64 factor antisera.

F.A.Q.

Click the question to get your answer

Antibodies can be frozen, but the process of freezing and thawing is damaging for the antibodies, and the titer will drop (even though not dramatically) due to decomposition.
We have not made further studies on this, and cannot guarantee the stability of the sera, but we have experienced that mostly the titer stays at the same level if a serum only is frozen and thawn once.

Yes, it is possible, if you place an order in advance with the expected total volume for the period. In return we guarantee to keep the reserved lot for you.

Depending on the needed volume, we might be able to develop the requested product. It can be anything from a new pool composition to a new serum.
